چکیده

Both assays are accepted reference methods for measurement of lactate dehydrogenase (LDH) activity (2-4). At pH 7.4, pyruvate -* lactate (P -* L) activity is about three times lactate -+ pyruvate (L P) activity, because equilibrium favors formation of lactate. Normal range for the P -* L assay is 130-265 spectrophotometric units at 25#{176} (3); normal range for the L P assay is 40-98 units at 25#{176} (5). The ratio obtained by dividing P L activity by L -* P activity varies for different serum specimens (4, 6). We have termed this the “P:L ratio”:

Optimal conditions for assaying human lactate dehydrogenase by the lactate-to-pyruvate reaction: Arrhenium relationships for lactate dehydrogenase isoenzymes 1 and 5.

Optimal reaction conditions to sassay human lactate dehydrogenase (lactate-to-pyruvate) were established for isoenzymes 1 and 5 at 25, 30, and 37 degrees C in diethanolamine and 2-amino-2-methyl-1,3-propanediol. Different substrate concentrations are required at each temperature. The conditions permit measurement of lactate dehydrogenase 1 and 5 with the lowest substrate concentrations that all...

Changes in lactate dehydrogenase, LDH isoenzymes, lactate, and pyruvate as a result of feeding low fat diets to healthy men and women.

A study was conducted to evaluate the effects on blood lipids and lipoproteins of feeding 21 healthy volunteers, 40-60 yr old, foods commonly eaten in the United States for two 40-day periods. Activities of lactate dehydrogenase (LDH) and LDH isoenzymes, lactate, and pyruvate were monitored. Results showed that LDH activity was significantly lower in all subjects at the end of the 25% fat-calor...

Measurement of total lactate dehydrogenase activity.

Lactate dehydrogenase (LD: EC 1.1.1.27) is the most important clinically of several dehydrogenases occurring in human serum. Lactate dehydrogenase is cytoplasmic in its cellular location and in any one tissue is composed of one or two of five possible isoenzymes. While many of its clinical applications involve quantification of one or more specific serum isoenzymes, an estimate of total LD is r...

How accurate are lactate dehydrogenase isoenzyme estimations by the thin-layer agarose fluorescent technique?

We offer an assessment of the accuracy of the thin-layer agarose fluorescent technique of Elevitch et al. [Am. J. Clin. Pathol. 46, 692 (1966)]. We used semi-purified human lactate dehydrogenase isoenzymes 1 and 5. Both the lactate lead to pyruvate and pyruvate lead to lactate assays [Clin. Chem. 20, 1462 (1974)] appear to give, within the errors of the techniques used, a substantially unbiased...

Optimum reaction conditions for human lactate dehydrogenase isoenzymes as they affect total lactate dehydrogenase activity.

Optimum reaction conditions at 30#{176} ± 0.5#{176} for two continuous spectrophotometric assay procedures, lactate to pyruvate (LP) and pyruvate to lactate (PL), were determined with respect to pH at 30#{176} (pH30) substrate concentration, and coenzyme concentration for the human LDH isoenxymes. For the P L procedure, broad pH30 optima were within the range of 7.20-7.40 for all the LDH isoenz...
